    IHC staining of anti-human CD15 recombinant (clone QA19A61) Spark Violet™ 423 on formalin-fixed paraffin-embedded human spleen tissue. After antigen retrieval using 1X Citrate Buffer (Cat. No. 420902), the tissue was incubated with 10 µg/mL of anti-human CD15 recombinant (clone QA19A61) Spark Violet™ 423 (blue) and anti-human CD8 (clone C8/144B) Alexa Fluor® 647 (red) at 4°C overnight. Nuclei were counterstained with Helix Green (green) (Cat. No. 425303). The image was captured with a 10X objective.

Description

CD15 is 3-fucosyl-N-acetyllactosamine (3-FAL), also known as Lewis X, 3-FAL, X-hapten, and SSEA-1. CD15 is expressed on granulocytes and monocytes. It has also been shown to be expressed on Langerhans cells and some malignant cells. CD15 has been implicated in adhesion, as well as chemotaxis, phagocytosis, and bactericidal activity.

Preparation

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is 5 µL per million cells in 100 µL staining volume or 5 µL per 100 µL of whole blood. For immunohistochemistry on formalin-fixed paraffin-embedded tissue, a concentration of 10 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

* Spark Violet™ 423 has a maximum excitation of 400 nm and a maximum emission of 415 nm.
