    HeLa cells untreated (panels A and B) or treated with 200 µM of CoCl2 (panels C and D) were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with Triton X-100 for 10 minutes, and blocked with 5% FBS for 1 hour. Cells were then intracellularly stained with 2.5 µg/mL of Alexa Fluor® 647 rat IgG2a, κ isotype control (panels A and C) (Cat. No. 400526), or with 2.5 µg/mL of Alexa Fluor® 647 anti-HMOX1 (clone W19398C) (Red, panels B and D) overnight at 4°C. Nuclei were counterstained with DAPI (Cat. No. 422801), and actin was counterstained with Flash Phalloidin™ Green 488 (Cat. No. 424201). Images were captured with a 40X objective. Scale bar: 50 µm

Description

HMOX1 is a member of the heme oxygenase family. Unlike its constitutive counterpart, it is an inducible enzyme triggered by external stimuli such as nitric oxide, growth factors, heavy metals, UV radiation, phorbol esters, inflammatory cytokines, heme, etc. Transcription of HMOX1 in tissue has been shown to be a product of the nrf2/ARE pathway where an increase in its protein levels results in heme degradation. The tissue protective effects of HMOX1 are a result of the bioactive products produced after heme degradation: iron ions, carbon monoxide, and biliverdin which is converted to bilirubin via biliverdin reductase.

Recommended Usage

Each lot of the test-sized antibody is quality control tested by intracellular immunofluorescent staining with flow cytometric analysis. For flow cytometric staining using the test sizes, the suggested use of this reagent is 5 µl per million cells in 100 µl staining volume or 5 µl per 100 µl of whole blood. It is recommended that the reagent be titrated for optimal performance for each application.

Each lot of the µg-sized antibody is quality control tested by immunocytochemistry. For immunocytochemistry using the ug size, a concentration range of 1.25 - 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application. 

* Alexa Fluor® 647 has a maximum emission of 668 nm when it is excited at 633 nm / 635 nm.

Application Notes

Clone was tested for ICC using three fix/perm methods (PFA/Triton-X, PFA/MeOH, MeOH only), the best method was the use of PFA/Triton X-100.

In the CoCl2 treated cells for ICC, staining was seen in less than 5% of cells.

Clone was tested for ICFC using cytokine fix/perm method only (based on ICC results).
