Whole cell extracts (15 µg total protein) from HEL cells untreated or treated with HSCF (50 ng/mL, 5 minutes), were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 1 µg/mL of purified anti-CD117 Phospho (Tyr703) (clone A22001B) (left panel) or anti-CD117 Pan (right panel) for 2 hours. Proteins were visualized by chemiluminescence detection using HRP Goat anti-mouse IgG (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. Description

Proto-oncogene CD117/c-KIT is a type III receptor tyrosine kinase that binds Stem Cell Factor (SCF). CD117 is used to identify certain types of hematopoietic progenitors in bone marrow, such as hematopoietic stem cells (HSC), multipotent progenitors (MPP), and common myeloid progenitors (CMP). Expression of CD117/c-KIT is also found in stem cell precursors from prostate, liver, and heart. CD117/c-KIT signaling plays a role in cell survival, proliferation, and differentiation; hence, its activation is required for melanocyte survival, hematopoiesis and gametogenesis. Aberrant activation of CD117/c-KIT due to gene mutations has been associated with gastrointestinal stromal tumors, testicular seminoma, mast cell disease, melanoma, acute myeloid leukemia, and others, while loss of activation is associated with piebaldism.

c-KIT is a 976 amino acid protein with a predicted molecular weight of 110 kD. It is a type III receptor tyrosine kinase protein that is composed of five immunoglobulin-like domains on the extracellular side and an intracellular tyrosine kinase domain.

c-KIT is a transmembrane protein but can also be found in cytoplasmic vesicles after internalization.
