The GeoMx® Digital Spatial Profiler (DSP) from NanoString enables researchers to perform spatial analysis of protein and RNA targets in FFPE and fresh frozen human and mouse samples. An important part of the GeoMx® DSP workflow is the identification of Regions of Interest (ROIs) in the tissue sample. The UV photo-cleavable oligo barcodes from probes attached to protein and RNA targets within these ROIs are then released and detected to create a detailed proteogenomic profile of the sample.

 

 

 

A robust method of selecting such ROIs involves the use of our fluor-conjugated antibodies as morphology markers in tissue samples to label tissue compartments and cell types. The morphology of the sample then guides the selection of ROIs based on biologically relevant features. Using the integrated segmentation algorithm in DSP software, ROIs may be automatically divided into discrete biological compartments or areas of illumination (AOI) for precise profiling and used to answer specific biological questions.
