Whole cell extracts (15 µg protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of purified anti-CDK1 (clone A20004C) overnight at 4°C.     ICC staining of purified anti- CDK1 (clone A20004C) on HeLa cells. The cells were fixed and permeabilized with ice-cold methanol, and blocked with 5% FBS for 1 hour at room temperature. The cells were then stained with 10 µg/mL of the primary antibody, followed by incubation with 2.5 µg/mL of Alexa Fluor® 594 Goat anti-mouse IgG (Cat. No. 405326) for 1 hour at room temperature. The image was captured with a 60X objective.

No. 928502), the tissue was incubated without (panel A) and with (panel B) 5.0 µg/mL of primary antibody followed by incubation with Alexa Fluor® 647 goat anti-mouse IgG (Cat. Description

Cyclin-dependent kinase 1 (CDK1) is a serine/threonine kinase that is ubiquitously expressed in eukaryotes. CDK1 is a catalytic subunit of a protein kinase called the M-phase promoting factor that universally induces entry into mitosis in eukaryotes. CDK1 has been shown to interact with numerous proteins including cyclin A1, cyclin A2, cyclin B1, cyclin B2, BRCA1, dynamin 2, Fyn, Lyn, and p53. In addition to regulation by protein-protein interactions, CDK1 is controlled by phosphorylation, particularly at conserved tyrosine residues. In humans, CDK1 is encoded by the gene CDC2 (cell division control protein homolog 2).

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 5.0 - 10.0 μg/mL is recommended. For immunoprecipitation, the suggested use of this reagent is 2.0 µg/test. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This product was tested and did not produce a detectable signal for mouse cross-reactivity in WB.
