Gated on CD45+ cells. A single cell suspension from C57BL/6 spleen was prepared and heat killed to isolate live cells using the MojoSort™ Mouse Dead Cell Removal Kit. Cells were stained with anti-mouse CD45 (Clone 30-F11) APC, Helix NP™ Blue and Apotracker™ Green. -
Representative live cell recovery comparison between competitor product A and MojoSort™ Mouse Dead Cell Removal kit. Dead cells were magnetically separated from heat-stressed C57BL/6 murine splenocytes (containing 40% dead cells) using recommended protocols for each kit. Competitor A required use of a specialized binding buffer containing high Ca2+ concentration. % live cell yield was calculated based on recovery of CD45+ Helix NP™ Blue- Apotracker™ Green- population relative to the unsorted population. Data represents compiled data from two technical replicates. -

MojoSort™ Mouse Dead Cell Removal Kit effectively removes dead cells to enrich mouse samples with high cell viability for downstream applications without using a specialized buffer containing high Ca(2+) concentration. It utilizes a complex formed between Apo-Monomer and Streptavidin nanobeads, which bind to phosphatidylserine residues exposed on the surface of apoptotic and dead cells with high affinity. Dead cells remain on the magnetically bound fraction and are depleted using magnetic separation. The untouched live cells are collected by decanting the liquid into a clean tube. These are the cells of interest; do not discard the liquid. Some of the downstream applications include functional assays, gene expression, phenotypic characterization, etc.
MojoSort™ reagents are also compatible with column-based cell separation systems available from other vendors. Optimized protocols for cell separation using columns from in-house testing are provided for each kit under the “Related Protocols” section, as well as representative data on the product webpage (where available). Data generated using column separators are indicated on the figure legend.
Due to the property of the beads, MojoSort™ reagents typically require dilution for optimal use on column separators. Where available, recommended dilution factors for each kit component based on in-house testing are provided under the “Application Notes” section of the webpage.

No, not currently. We have found that cells are functional without the need to detach the magnetic Nanobeads.

- Are MojoSort™ Nanobeads compatible with other commercially available magnetic separation systems?

MojoSort™ magnetic particles can be used with other commercially available magnetic separators, both free standing magnets and column-based systems. Because MojoSort™ protocols are optimized for the MojoSort™ separator, the protocols may need to be adjusted for other systems. Please contact BioLegend Technical Service for more information and guidance. We do not recommend using MojoSort™ particles for BD’s IMag™ or Life Technologies’ DynaMag™.
