Following antigen retrieval using 1X Citrate buffer 1X pH 6.0 (Cat. No. 420902). The tissue was incubated with 10 μg/mL of Spark Violet™ 423 anti-human CD15 (SSEA-1) (clone HI98, blue) and Alexa Fluor® 647 anti-human IgD (clone W18340A, red) overnight at 4°C. Nuclei were counterstained with Helix green (green) (Cat. No. 425303). Images were captured with a 10X objective.

Description

CD15 is 3-fucosyl-N-acetyllactosamine (3-FAL), also known as Lewis X, 3-FAL, X-hapten, and SSEA-1. CD15 is expressed on granulocytes and monocytes. It has also been shown to be expressed on Langerhans cells and some malignant cells. CD15 has been implicated in adhesion, as well as chemotaxis, phagocytosis, and bactericidal activity.

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is 5 µL per million cells in 100 µL staining volume or 5 µL per 100 µL of whole blood. For immunohistochemistry, a concentration of 10 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

* Spark Violet™ 423 has a maximum excitation of 400 nm and a maximum emission of 415 nm.

Clone HI98 has been described to specifically react with 3-fucosyl-N-acetyllactosamine (3-FAL), also called X-hapten, SSEA-1, CD15, or Lewis X. Additional reported applications (for the relevant formats) include: immunohistochemical staining of acetone-fixed frozen sections5 and formalin-fixed paraffin-embedded tissue sections.
