Description

Plasminogen activator, also known as PA, is a serine protease which converts inactive proenzyme plasminogen to active protease plasmin. PA has been classified in two types: urokinase–like activator and tissues plasminogen activator (t-PA). The principal plasminogen activator is t-PA, which is synthesized mainly by endothelial cells, and is secreted locally after stimulation of the endothelium by histamine, adrenalin, thrombin, FXa, and hypoxia. T-PA is secreted as a protein of 527 amino acids. The molecule possesses four highly conserved structural domains: the amino-terminal fibronectin type-1 domain, an epidermal growth factor-like domain, two kringle domains, and the carboxi-terminal which contains the serine protease catalytic domain. The concentration of t-PA in serum is approximately 0.1 nM under normal physiological conditions, and a big portion of t-PA in serum is associated to plasminogen-activator inhibitor type 1 (PAI-1). The inhibitor interacts with positively charged amino acids near the catalytic site of t-PA. The t-PA-PAI-1 inactive heterodimeric complexes are cleared from the circulation by the liver, while the complexes are internalized by the low density lipoprotein receptor-related protein (LRP-1). T-PA plays an important role in the removal of intravascular fibrin deposits, and it has been used in the treatment of acute ischemic stroke.

Human tPA, amino acids (Ser23-Pro562) (Accession# NM_000930), with a C-terminal 6His tag was expressed in 293E cells.
Molecular Mass
The 564 amino acid recombinant protein has a predicted molecular mass of approximately 61.4 kD. The DTT-reduced and non-reduced protein migrates at approximately 40-45 kD and 65 kD respectively by SDS-PAGE.
Purity
>95%, as determined by Coomassie stained SDS-PAGE.
Formulation
0.22 µm filtered protein solution is in MES, pH 5.5, and in 0.5 M NaCl.
Endotoxin Level
Less than 1.0 EU per µg of protein as determined by the LAL method.
Concentration
10 and 25 µg sizes are bottled at 100 µg/mL.

Unopened vial can be stored at -70°C for six months. For maximum results, quick spin vial prior to opening. Avoid repeated freeze/thaw cycles.

Human t-PA cleaves the peptide substrate Bz-GGR-AMC with a specific activity value higher than 300 pmol/µg/min.

Distribution

It is produced mainly by endothelial cells. It is found in the extracellular matrix (ECM) of most animal tissues and fluids such as urine and blood.

Converts plasminogen into plasmin, cell migration, angiogenesis, wound repair, inflammation, immunity, and tumor metatases. It is regulated by plasminogen activator inhibitor type 1 or neuroserpin.

What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.

Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.

Does specific activity of a recombinant protein vary between lots?

Specific activity will vary for each lot and for the type of experiment that is done to validate it, but all passed lots will have activity within the established ED50 range for the product and we guarantee that our products will have lot-to-lot consistency. Please conduct an experiment-specific validation to find the optimal ED50 for your system.
