Total lysates (15µg protein) from HeLa (Lane 1), 5nM (Lane 2)and 20nM (Lane 3) UQCRC1 siRNA treated HeLa cells were resolved by electrophoresis (4-20% Tris-Glycine gel), transferred to nitrocellulose, and probed with purified anti-UQCRC1 antibody (upper) or GAPDH antibody (lower). Proteins were visualized using an HRP Goat anti-mouse IgG Antibody or HRP Donkey anti-rabbit IgG Antibody and chemiluminescence detection. Lane M is the molecular weight ladder.

    HeLa cells were fixed with 4% paraformaldehyde (PFA) for fifteen minutes, permeabilized with 0.5% Triton X-100 for three minutes, and blocked with 5% FBS for 60 minutes. Then the cells were stained with DAPI (blue) only (left) or DAPI and intracellular anti-UQCRC1 antibody (right, clone O91G2 at 2 µg/ml) overnight at 4°C followed by Alexa Fluor® 594 (red) conjugated goat anti-mouse IgG for one hour at room temperature. The image was captured with a 60X objective.

Description

The ubiquinol-cytochrome c reductase complex, also called mitochondrial complex III, is the third complex in the mitochondrial electron transport chain. The complex III is composed of 11 subunits that form a multi-segment transmembrane protein. It is part of the mitochondrial respiratory chain and plays essential roles in the synthesis of ATP. UQCRC1 may mediate formation of the complex between cytochromes c and c1.

The dysregulated UQCRC1 is involved in impaired mitochondrial electron transport chain function and was found in clear cell renal cell carcinoma and Alzheimer's disease.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 1.0 µg per ml.For immunocytochemistry, a concentration range of 0.5 - 5.0 μg/ml is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
